G-四链体DNA对于癌症治疗的研究综述

摘要:
c-myc肿瘤基因的过表达是癌症启动和发展的分子标志。针对c-myc基因的研究是一种合理且有效的癌症治疗策略。一种特殊的DNA二级结构——G-四链体DNA在核酸酶超敏元件Ⅲ1(NHE Ⅲ1)区域内形成,该区域位于c-myc基因的P1启动子上游,驱动其大部分转录。近年来,对于G-四链体结构的靶向研究一直是抗癌治疗的焦点。本篇综述,首先概述了G-四链体结构的发现及其在体外和细胞内形成的证据,然后描述了G-四链体在调节c-myc基因表达中的功能作用,总结了几种作用于G-四链体结构的小分子物质,最后总结了它们作为新型抗癌药物的潜力。

Abstract: The overexpression of c-myc tumor genes is amolecular marker of cancer initiation and development.Research on c-myc gene is a reasonable and effective cancer treatment strategy.A special DNA secondary structure-G-four chain DNA in nuclease super sensitive element Ⅲ 1 (NHE Ⅲ 1) formed in the area,the area is located in the c-myc gene P1 promoter upstream, driving most of its transcription.In recent years, the target research of G-four-chainstructure has been the focus of anticancer therapy.In this review, we first reviewed the discovery of the G-quadruplet structure and its in vitro and intracellular evidence, then described the functional role of g-quadruplet in regulating c-myc gene expression, summarized several small molecular substances that act on the G-quadruplet structure, and finally summarized their potential as novel anticancer drugs.
