HPLC法同时测定泽兰中迷迭香酸与白桦酸的含量
摘 要
目的:泽兰在我国资源较为丰富,地域分布较为广泛。由于中国药典中并未收有关泽兰含量标准的数据,因此本实验将利用高效液相色谱技术对泽兰中含量较多的迷迭香酸与白桦酸同时进行含量测定,为药典中泽兰的含量标准提供参考。
方法:本实验利用甲醇加热回流法对泽兰进行提取,应用硅胶柱层析法、薄层层析法分离纯化出两种单体化合物迷迭香酸和白桦酸,采用反相硅胶色谱柱:Thermo C18 色谱柱 (4.6 mm×250 mm, 5 μm);以0.2 % 磷酸水溶液(A)-乙腈(B)为流动相进行梯度洗脱;检测波长:210 nm;柱温:35 ℃;进样量:10 μL;流速:1 mL/min的条件从泽兰甲醇提取物中测得迷迭香酸和白桦酸的百分含量。
结果:在泽兰甲醇提取物中,迷迭香酸在0.001 mg/mL-0.03 mg/mL范围内的线性关系良好(R2 = 0.9996)平均回收率为98.21 %;白桦酸在0.02 mg/mL~1 mg/mL范围内的线性关系良好(R2 = 0.9997)平均回收率为98.17 %。方法学考察中精密度、重复性、稳定性、加样回收率的RSD值均小于2 %,最后测得泽兰乙醇提取物中迷迭香酸的含量为22.880 mg/g,白桦酸的含量为73.554 mg/g。
结论:利用高效液相色谱技术同时测定泽兰甲醇提取物中的迷迭香酸和白桦酸的含量,该方法可靠可行,重复性好、精密度高、稳定性强,可为药典泽兰的质量标准提供参考,同时可为泽兰的进一步研究奠定基础。

Results: The linear range of rosmarinic acid was 0.001 mg/mL-0.03 mg/mL (R2 = 0.9996).The average recovery rate of rosmarinic acid was 98.21 %.The linear range of betulinic acid was 0.02 mg/mL to 1 mg/mL (R2 = 0.9997).The average recovery was 98.17 %.The RSDs of precision, reproducibility, stability and recovery were all less than 2 %.Finally, the contents of rosmarinic acid and betulinicacid in ethanol extract of Zanthopanthus alba were 22.880 mg/g and 73.554 mg/g respectively.
Conclusion: The method for simultaneous determination of rosmarinicacid and betulinic acid in the methanol extract of Zetanthus officinalis was developed by high performance liquid chromatography (HPLC). The method was reliable, feasible, accurate and stable,and could provide a reference for the quality standardof Zetanthus officinalis and lay a foundation for the further study of Zetanthus officinalis.
